Yes, if you solder the missing IC (MCP23017) the additional GPIO functionality will be restored. You don’t need any additional steps beyond using it in the code.
When soldering, be careful not to damage the e-paper panel with high temperatures. It is best to solder the IC using a soldering iron without using hot air.
