I want to use an Inkplate screen as a digital clock with seconds. If I perform partial refreshes every second and a full refresh every 6 hours, how long would this screen last? I had a Waveshare screen running this pattern that eventually failed around 1.5 years.

That’s quite a stress test! You’ll experience ghosting with a full refresh only every 6 hours. We recommend a full update every 50 or so partial updates to remove the ghosting. Your mileage may vary.
Regarding durability, it’s hard to say due to the analogue and chemical nature of e-Paper. We’ve heard of projects lasting 3+ years, but that’s with way less frequent updates, which is more suited for e-Paper. The good thing is, even if the e-Paper fails, it’s easily replaceable and you can continue using the project.
