Does the API allow me to increase the clock speed of the I2S system to 2MHz to operate a MEMS microphone in the Ultrasound Region?

The onboard I2S can go up to 150 MHz so it should work with 2 MHz no problem. You will have the pins out for I2S and then you can use HAL functions within Arduino to implement anything you need from the pins. We are also implementing an I2S master functionality natively through the library, so that should be easy to use. For anything else, you can use HAL as it’s included in STM32duino which we use as the basis for our board definition.
